It comprises three subsystems (fluidic, optical, and electronic) organized into two interconnected units. The fixed alignment and “cuvette-based” technology ensure reliability and good signal resolution. The system acquires, sorts, and analyzes cells in a liquid suspension labeled with a fluorescent dye. The high degree of automation and the guided procedures available with BD FACSMelody facilitate the achievement of consistent results, without necessarily requiring operators with a high level of experience. The instrumentation is placed within a dedicated area equipped with a Class II Biosafety Cabinet, ensuring the correct execution of experiments. Features: EXCITATION SOURCE The instrument can detect up to 11 parameters simultaneously (9 fluorescences + FSC + SSC). Three lasers are installed, spatially separated: Blue 488 nm (20 mW), Red 640 nm (40 mW), Violet 405 nm (40 mW). ALIGNMENT The presence of spatially separated lasers ensures low background noise and low compensations. OPTICS The instrument is equipped with a Reflection Optical System. MINIMUM PARTICLE DETECTION SIZE Thanks to its high dimensional resolution and high sensitivity to fluorescence parameters, the BD FACSMelody instrument can easily distinguish fluorescent particles with a diameter of 100 nanometers (0.1 microns) from background noise. ELECTRONICS Completely digital for signal processing and data acquisition. Up to 40,000 events/second can be acquired on eleven parameters. Resolution: 262,144 channels on all parameters with digital sampling. COUNTING CHAMBER In gel-coupled quartz, designed to minimize background noise and ensure maximum sensitivity.
